The laboratory is N005 room on the 1st basement floor of the General Research Building. We have equipment that allows you to make and observe DNA circuits.

[Basic laboratory equipment]

Basic experimental work such as buffer and DNA solution preparation and electrophoresis experiments is performed.

[Ultraviolet spectrophotometer] V730 (JASCO Corporation)

Since the absorbance of a DNA strand changes depending on its structure and state, it is used when analyzing the structure of the designed DNA strand.

 

[Fluorescence spectrophotometer] FP-8300 (JASCO Corporation)

It is used to verify the operation of DNA circuits. By modifying a specific DNA strand with a fluorescent probe, the time evolution of concentration change can be measured as fluorescence intensity. You can also use a temperature controller to verify the operation of the DNA circuit at the desired temperature. It is the most frequently used measuring instrument in our laboratory.

[Plate Reader] Spectra Max iD3 (Molecular Device)

It is used to verify the operation of DNA circuits together with a fluorescence spectroscopic altimeter. The feature of this device is that it is possible to measure under 96 kinds of conditions at the same time by using a 96-well plate. In addition, it is equipped with various measurement modes such as fluorescence measurement and absorbance measurement.

 

[Automatic dispensing device] epMotion 5073 (eppendorf)

In order to experiment with the operation of DNA circuits, it is necessary to prepare multiple types of designed DNA strands in microtubes at appropriate concentrations. This work is called pipetting and is done manually. For example, preparing a sample on a 96-well plate under 96 different experimental conditions would be a daunting task. In this device, the robot automatically performs manual pipetting. It is an excellent tool that can perform many sample preparation tasks quickly and accurately.
